IRIS Dyes labelled nucleotides can be used as labels for DNA and RNA with the most widely known methods such as nick translation, random priming, polymerase chain reaction, cDNA synthesis etc.
The fluorescent analogs are purified by HPLC chromatography. Analytical HPLC is used as a quality control check to ensure chemical purity >95%. UV/VIS absorption spectra are obtained in aqueous phosphate buffer to determine concentration.
IRIS dyes labelled dUTP, dCTP, UTP and CTP can be enzymatically incorporated into probes for use in many molecular biology applications. The use of fluorescent nucleotides with different fluorescence emission profiles allows multicolor applications. We have developed the following protocols for incorporating these fluorescent nucleotides into hybridization probes. Depending on your application, some further optimization of the reaction conditions may be necessary.
